- Dean goes to Chicago with the demon Crowley to meet Death, one of the Four Horsemen of the Apocalypse, and take his ring, so they can try to stop Lucifer.
- Sam & Bobby & Castiel try to stop the shipments of the deadly Croatoan-virus-infected vaccine.
- Dean agrees that he won't try to stop Sam from taking on Lucifer and fighting him.
- They go to Detroit and Lucifer takes over Sam's body with very little if any struggle.
The title of the episode is drawn from an Iron Maiden song, "2 Minutes to Midnight".
